Semaglutide Extends Lifespan in Older Mice, Raising New Questions About Aging

A new mouse study suggests semaglutide, a GLP-1 drug used for weight loss, may slow aging and extend lifespan.

A new study is adding an unexpected possibility to the growing list of effects linked to GLP-1 drugs such as semaglutide. Researchers gave the drug to female mice that were already 20 months old and followed them for the rest of their lives. Their median lifespan rose from 742 days in untreated mice to 834 days in the semaglutide group, an increase of about 12 percent. The researchers also looked beyond lifespan. Treated mice showed better performance in tests of movement, exploration and memory, while their tissues showed fewer signs of age-related inflammation and damage. In a separate experiment, the researchers compared semaglutide with calorie restriction, a well-studied approach that can extend lifespan in animals. The drug produced several benefits similar to reduced food intake and appeared to provide additional improvements in areas such as memory and blood-sugar control. That does not mean semaglutide has been shown to extend human life. The lifespan result comes from an animal study, and the researchers are still working to determine exactly why the drug produced these effects. The findings are especially interesting because GLP-1 medicines already affect several systems involved in metabolism, inflammation and cardiovascular health. Other 2026 research has also reported early evidence that semaglutide can influence biological aging markers in humans, but that evidence is not the same as proving longer human lifespan. The bigger question is whether GLP-1 drugs are simply reproducing some benefits of eating less or whether they also affect aging through additional biological pathways. The mouse study suggests there may be more going on, but answering that question will require further research in humans.
